Evaluation of Different Molecular Markers for Genotyping Non-Saccharomyces Wine Yeast Species

Wine quality is determined by the particular yeast strains prevailing at various stages of fermentation. Therefore, ability to make an easy, fast, and unambiguous discrimination yeasts strain level great importance. Here, tandem repeat-tRNA (TRtRNA) method with 5GAC or ISSR-MB primer sets random amplified polymorphic DNA (RAPD) analysis (GTG)3, R5, RF2 oligonucleotides were tested on non-Saccharomyces wine species. The TRtRNA-PCR employing showed highest capacity in discriminating Lachancea thermotolerans Metschnikowia pulcherrima isolates. RAPD was most efficient resolving Starmerella bacillaris isolates, although it produced few bands. R5 discriminate among Issatchenkia orientalis, Hanseniaspora guilliermondii, Pichia anomala either exhibited Torulaspora delbrueckii (GTG)3 for H. uvarum Here we concluded that both RAPD-PCR offer rapid means typing However, each performs better a given species when paired set. present results can be useful research fast fingerprinting yeasts.
